Do home folders and the homes shared folder both take up space on my NAS?

No, despite the appearance of being located in different locations, the folders contain the same content and don't take up any additional space. 

Enabling home folders creates a personal folder for each local and domain user on the NAS. All user home folders are located in the homes shared folder. By default, only administrators can access this parent directory, but each user is able to access their assigned subfolder. To each user, the home folder is named home, but within the homes folder, administrators see the folder listed with the user's name.

For example, let's imagine a user (Example_User) on your NAS. To this user, the home directory appears to be located at ~/home. To an administrator, the directory is located at /homes/Example_User.
